- Abstract: Objetivo: The aim of this study was to evaluate the effect of physical preconditioning on the reactivity of mesenteric artery from rats submitted to a pulmonary ischaemia-reperfusion (IR). Métodos e Resultados: Wistar male rats were divided into sham operated (SHAM); IR sedentary (IRSD) and IR trained (IRTR) groups. Run training (RT) was performed for 5 days/week, each session of 60 min during 8 weeks at intensity of 66% of VO2max. After RT, the animals were submitted to pulmonary IR for 90 min and reperfusion for 2 hours. Rats were sacrificed and the mesenteric artery ring was isolated. Concentration-response curves to acetylcholine (ACh), histamine (HIST), and phenylephrine (PHE) were obtained. IR reduced the potency for ACh (SHAM: 7.65±0.12; IRSD: 7.00±0.05; IRTR: 6.88±0.06) and for HIST (SHAM: 5.26±0.05; IRSD: 5.05±0.08; IRTR: 5.52±0.02). Neither potency nor Emax were altered for PHE. RT did not modify the potency for ACh, but produced a significantly increase of potency for HIST, about 3-fold. No changes in Emax were observed for ACh and HIST. Conclusões: Physical preconditioning for 8 weeks did not prevent the impairment of sensitivity of mesentery artery ring for ACh. In contrast, a super sensitivity of relaxing response for HIST was observed in mesenteric ring from trained groups
