Effect of bay 41-2272 in the pulmonary hypertension induced by hypoxia in anaesthetized beagles (2005)

- Abstract: Objetivo: BAY 41-2272 has been reported as a novel NO-independent activator of the soluble guanylate cyclase. BAY 41-2272 relaxes aortic rings in vitro and reduces mean arterial blood pressure in normal and hypertensive rats. In this study, we investigated whether BAY 41-2272 counteracts the pulmonary hypertension (PH) induced by hypoxia in anaesthetized dogs. Métodos e Resultados: 12 male Beagles dogs (8-14 kg) were anaesthetized with pentobarbital sodium (30 mg/kg, i.v), and supplemented with fentanyl (10 µg/kg/h) and diazepam (700 µg/kg/h). In the hypoxia group, dogs were submitted to a 12% FiO2. The following parameters were evaluated: blood pressure (BP), heart rate (HR), PH, cardiac index (CI), index of systemic and pulmonar vascular resistance (ISVR), (IPVR). Hypoxia caused a marked PH (at 5 min: from 7.7±1.8 to 21.8±3.3 mmHg; P<0.01). This was accompanied by increases in CI (from 3.3±0.4 to 4.7±0.5 mmHg) and IPVR (125±12 to 250±32 D.s/cm5/m2). BAY 41-2272 (10 µg/kg/min, i.v.) prevented PH and the increases in IPVR and IC. Conclusões: Our results suggest prolonged stimulation of sGC in a NO-independent manner by BAY-412272 counteracts the vasoconstrictor mechanisms triggered by hypoxia at the pulmonary bed
