- Abstract: Objective: Schwann cells (SC) produce and release neurotrophic factors that induce the survival and regeneration of neurons following nerve injury, a process that may be influenced by the local release of inflammatory cytokines. The present study examined the effect of SC conditioned medium (SCCM) cultured in the presence of transforming growth factor beta 1 (TGF-'beta'1) or tumor necrosis factor alpha (TNF-'alfa') on fiber growth of brainstem neurons in vitro. Material and Methods: Primary neuronal cultures were established from neonatal Wistar rats while SC cultures were obtained from sciatic nerves explants of adult rats. SC were treated or not with TGF-'beta'1 or TNF-'alfa' for 24 hours. Neuronal cells received the SCCM every 12 hours during 96 hours. As controls, neurons received neuronal or SC culture medium. The areal fraction (AA) of neuronal fibers per neuron was quantified by means of stereologycal method at 24, 48 and 72/96 hours after treatment under contrast-phase microscopy. Results: The SC medium (control) induced fiber sprouting through out periods of analysis. However, the SCCM induced further sprouting only at 72/96 hours compared to SC medium. At the time, the effect of SCCM was not altered when SC cultures were pre-treated with TNF-'alfa' or TGF-'beta'1. Conclusion: The results showed that SCCM may stimulate the neurite outgrowth of brainstem neurons, a phenomenon that is not potentiated by TNF-'alfa' or TGF-'beta'1
