Endogenous angiotensin modulate hindlimb resistance reflex control in spontaneously hypertensive and normotensive rats (2004)

- Abstract: Objetivo: We sought to investigate the effects of chronic AT1 receptors blockade on reflex control of hind limb resistance in hypertensive and normotensive animals. Métodos e Resultados: Male SHR and WKY, aged 3 month and treated orally with vehicle (VEH=1ml/kg/day) or losartan (LOS=10 mg/kg/day) during 5 days, were chronically instrumented (day 3) with arterial/ venous catheters and iliac flow probe (Transonic). Arterial pressure (AP), heart hate (HR) and hindlimb blood flow (HLF) were recorded (and HL resistance calculated, HLR=MAP/HLF) in conscious freely moving rats on day 5. Reflex control of HLR was determined during loading/unloading of baroreceptors (graded doses of phenylephrine/sodium nitroprusside i.v).In both groups LOS reduced MAP (~13%) by reducing basal HLR (SHRLOS=67.0+-5.0 and WKYLOS=58.8+-10.7 mmHg/ml/min/g, -35% and -32% vs. respective VEH-treated groups) without changing HR and HLF levels. LOS caused leftward and downward displacement of HLR x MAP function curves. Conclusões: Endogenous angiotensin II, besides the tonic control of HLR and pressure in hypertensive and normotensive subjects, is also important to maintain both the reflex sensitivity and the high operational range for reflex control of skeletal muscle resistance. In addition data show that angiotensin II modulates (via AT1 receptors) not only reflex-induced vasocontriction, but also reflex-induced vasodilation during baroreceptors loading.
