Cyclosporin A attenuates skeletal muscle fiber diameter increase during regeneration of fast and slow-twitch muscles (2004)

- Abstract: Objetivo: Skeletal muscle has remarkable capacity to regenerate after injury. Previous study has demonstrated that the treatment with cyclosporin A (CsA), a pharmacological inhibitor of calcineurin, is able to inhibit skeletal muscle regeneration, suggesting that calcineurin is important for this process (Cell Tissue Res 308:143, 2002). However, in this study it has been studied the effects of CsA administration only in a slow-twitch muscle. In the present work we investigated the effect of calcineurin inhibitor, CsA, on macroscopic and histomorphometric aspects of regenerating slow and fast-twitch muscles induced by cryolesion. Métodos e Resultados: Wistar rats (n=12) weighing 230-300 g were treated with CsA for 5 days. On the 6th day, animals were submitted to cryolesion of the left soleus and tibialis anterior (TA) muscles and were sacrificed at 21 days after cryolesion. The muscles were removed, frozen and stored in liquid nitrogen. The results show that cryolesion did not alter the body weight gain of the animals after 21 days of regeneration (p= 0.001) and CsA was able to significantly reduce the body weight in the same period (p=0.01). CsA treatment and cryolesion individually and combined were able to decrease muscle weights of soleus (p<0.05) and TA (p<0.05). Furthermore, CsA treatment was able to decrease cross-sectional area of both soleus and TA muscles of cryoinjured animals, (2298±452 vs 764±439 mg; 2108±930 vs 792±640 mg; p<0.001; respectively).Histological sections of both soleus and TA muscles stained with toluidine blue revealed similar regenerative responses after cryolesion and CsA was able to significantly minimize such responses, i.e. centralized nuclei and split fibers. Conclusões: These results indicate that calcineurin plays a key role for regeneration of both slow and fast-twitch muscles
