Toxic effect of fatty acids on J774 cells (2004)

- Abstract: The immunomodulatory properties of fatty acids have been described for more than half a century. Since then, it has been shown that some of these e_ects are due to the toxic e_ects of the fatty acids. In this study, we evaluated the e_ect of increasing concentrations of palmitic, oleic, linoleic and eicosapentaenoic acids on J774 cells, a model of murine macrophages. The cells were treated for 24 hours in the presence of 25 to 300 _M of the fatty acids and analyzed by ow cytometry and uorescence microscopy. The following parameters were evaluated: morphological changes, membrane integrity, chromatin condensation, DNA fragmentation and mitochondrial transmembrane potential. All fatty acids tested promoted apoptosis and necrosis of the cells and this e_ect was dependent on the concentration of the fatty acids. We observed an increase in cell granularity, loss of membrane integrity, increase in DNA fragmentation and loss of mitochondrial transmembrane potential. Palmitic acid was the most toxic fatty acid tested, inducing membrane and DNA changes at 75 _M. Oleic acid had toxic e_ects at 100 _M, linoleic acid at 250 _M and eicosapentaenoic acid at 150 _M. These _ndings suggest that the toxic e_ect of fatty acids on J774 cells is dependent on the carbon chain length and the degree of insaturation
