Combined immunization prtocol based on priming with parenteral DNA vaccine and boosting with oral attenuated Salmonella strain confers protective immunity to enterotoxigenic Escherichia coli in a murine model (2003)

- Abstract: In this work we evaluated the immunogenicity and protective efficacy of combined priming/boosting protocol based on the structural subunit (CfaB) of the CFA/I fimbriae of enterotoxigenic Escherichia coli (ETEC), which mediates attachment of bacteria to the human intestinal epithelium, encoded by a DNA vaccine and a recombinant attenuated Salmonella Typhimurium aroA strain (HG3). BALB/c mice were i.m. immunized with 2 doses (100 µg/dose) of the DNA vaccine (pRECFA) and after 4 weeks orally boosted with 2 doses (1010 cfu) of the HG3 strain, with or without addition of (25 µg/dose) of a mucosal adjuvant (LTR192G). Our results show that animals submitted to the prime/boost vaccine regimen, either with or without the adjuvant, induced a clear synergic response both for systemic (IgG) and intestinal secreted (sIgA) antibodies. Moreover, sera of mice submitted to the vaccine consortium inhibited the adhesion of CFA/I+ bacteria as demonstrated by in vitro and in vivo tests carried out with Caco-2 cells and using a newborn mice challenge assay, respectively. CFA/I-specific IgA antibodies were also detected in colostrums ingested by newborn mice fostered by vaccinated DBA2 females. Finally, we detected complete (100%) protection to lethal challenge (2 x 107 cfu) with a virulent CFA/I+ ETEC strain given to newborn suckling mice following immunization of pregnant female mice with the complete priming/boosting protocol but not with each vaccine strategies alone. Thiswork shows that the DNA vaccine efficiently boosted mucosal antibody response following oral stimulation with attenuated Salmonella cells and represent a promising alternative for the development of new vaccine protocols against enteric pathogens
