Mycoplasma arthritidis superantigen (MAM) induces iNOS and COX-2 in murine peritoneal cells: modulation by PAF (2003)

- Abstract: Aims: We investigated whether MAM is able to induce cyclooxygenase-2 (COX-2) or inducible nitric oxide synthase (iNOS) expression and release of prostaglandin E2 (PGE2) or nitric oxide (NO) from murine macrophages and its modulation by PAF. Methods: Resident peritoneal cells from male C3H/HePas mice were incubated with MAM (1/100) in the presence or absence of PAF-antagonist (WEB 2170) or prostaglandin synthesis inhibitor (Nimesulide). The COX-2 or iNOS expression was determinated by immunoblotting analysis and PGE2 or NO levels by EIA or Griess reaction, respectively. Results: Following activation of peritoneal cells with MAM production of NO increased with incubation time whereas iNOS expression was maximal at 20 h, decreasing thereafter. The peak level of PGE2 was reached after 20h (150 ng/mL) whereas the peak of COX-2 expression was at 12h.The kinetics of iNOS expression was not altered by WEB2170 treatment, however, NO were significantly lower in the treated group (58 vs 78 microM, at 40h). COX-2 expression and PGE2 production were significantly lower (122 vs 150 ng/mL, at 20 h) in the WEB-treated group. NO production inhibited by WEB2170 treatment was reversed by nimesulide. Nimesulide abolished the PGE2 and increased NO production induced by MAM (26% increase). Addition of PGE2 to the cultures together with MAM, reduced NO production (27% decrease). Conclusions: The results show that MAM induces iNOS and COX-2 expression, andrelease of NO and PGE2 from resident peritoneal cells. Endogenous PAF seems to modulate MAM-induced NO and PGE2 production at the level of COX-2, but not iNOS expression
