Modulation of allergic and immune complex-induced lung inflammation by bradykinin receptor antagonists (2003)

- Abstract: Objective: The effect of antagonists of bradykinin B2 receptor, HOE-140 (constitutive) and of B1receptor, R-954 (inducible) was studied in allergic and immune-complex-induced lung inflammation. Methods and results: Lungs of BALB/c mice were examined 24h after induction of lung inflammation either allergic (ovalbumin-sensitized submitted to two aerosol of antigen, one week apart) or immune-complex induced (intratracheal instillation of IgG antibodies to BSA followed by intravenous antigen). Compounds HOE-140 or R-954 were given intraperitoneally (100 mug/kg), 30 min before induction. In allergic inflammation, pre-treatment with R-954 reduced eosinophil infiltration (92%), mucus secretion (75%) and the airway hyperreactivity (84%) to methacholine. Pre-treatment with HOE-140 increased eosinophil infiltration (207%) but did not affect the other parameters. In immune-complex inflammation, HOE-140 increased neutrophil infiltration (58%) but not their activation nor the hemorrhagic lesions. Pre-treatment with R-594 did not change the parameters examined.Conclusion: These results suggest that activation of the constitutive bradykinin receptor down-regulates both types of lung inflammation whereas the inducible receptor participates only in the allergic inflammation where it has pro-inflammatory effects
