High molecular weight components from ascaris suum suppress immune response to an unrelated antigen through IL-4 and IL-10 cytokines (2003)

- Abstract: Aims: The extract from Ascaris suum worms (Asc) stimulates a Th2-type response and impairs the immune response to ovalbumin (OA) in mice. IL-4/IL-10 cytokines are involved in the suppressive effect of Asc, because treatment of OA+Asc-immunized DBA/2 mice with anti-IL-4+anti-IL-10 mAb abolished the suppression of OA-specific Th1-related parameters. PI (high MW proteins from Asc), which are responsible for the suppressive effect, elicited IL-4/IL-10 production in large quantities. In contrast, PIII (low MW proteins), that stimulates IgE antibodies, induced a Th0-type response. Thus, we decided to work with PI apart from PIII to investigate the real participation of PI-induced IL-4/IL-10 in down-regulation of Th1/Th2 responses to OA. Materials: IL-10-/- mice, IL-4-/- mice treated with anti-IL-10 mAb or wild-type mice were immunized with OA or OA+PI in adjuvant.Eight days later, we compared the immediate and delayed-type hypersensitivity reactions (HRs) in these mice when challenged in the footpad with aggregated OA, the levels of OA-induced antibodies and IFN-gamma/IL-2 cytokines productionIn the absence of IL-10, the suppressive effect of PI on the HRs to OA was abolished. Total recovery of IFN-gamma/IL-2 levels in OA+PI-immunized mice was only obtained in the simultaneous absence of IL-10/IL-4. The suppression of OA-specific Ab prodproduction was also reduced in IL-10-/- mice (IgG1/IgE) and in IL-10-deficient, IL-4-/- mice (IgG2c). CONCLUSIONS: These results indicate that IL-10 is involved in the suppression of OA-specific Th2-related parameters by Ascaris high MW componentcomponents, while IL-4/IL-10 act together in the suppression of Th1 parameters.
