Involvement of TH14 protein from Saccharomyces cervisiae in mitochondrial stability and normal oxidative condition (2003)

- Abstract: Objectives: The Thi4 protein from S. cerevisiae is responsible for the thiazol production in thiamin (vitamin B1) biosynthesis and the thi4 mutant strain shows an increased frequency of colonies with mitochondrial dysfunction upon treatment with genotoxic agents and after growth at 37oC. To investigate the Thi4 involvement in maintaining mitochondrial stability, we analyzed the oxidative condition of thi4 strain under 37oC. Methods and Results: We measured the activities of Superoxide Dismutase (SOD), Catalase (CAT) and Glutatione Peroxidase (GPX), the concentration of biomarkers of protein oxidation and the efficiency oxygen consumption of thi4 mutant and wild type strains, after growth at 37oC. The activities of SOD and GPX were significantly increased in thi4 strain grown at 37oC.The levels of protein carbonyl groups were also increased, while those from thiol groups were decreased. The rate of oxygen consumption by the thi4 mutant was smaller than in wild type strain after 37oC growth in both YPD and in YPG. Conclusion: These results corroborate to the previously observed involvement of Thi4 in maintaining mitochondrial stability and metabolism, pointing to a putative role related to the control of redox status in yeast cells
