Biosynthesis of dolichol and ubiquinone isoprenic chain in Leishmania amazonensis (2003)

- Abstract: Sterols are essential for cell viability and proliferation. The sterol biosynthesis for most eukaryotes derives from the mevalonate pathway. From acetyl-coenzyme A as the main substrate, most organisms synthesize hydroxymethylglutaryl-CoA (HMG-CoA) which is reduced to mevalonate by HMG-CoA reductase. Mevalonate is converted into isopentenyl pirophosphate (IPP) which generates geranyl pyrophosphate and farnesyl pyrophosphate, the main precursors of polyisoprenoids such as dolichol, ubiquinone, cholesterol and prenyl groups transferred to prenylated proteins. The sterol biosynthesis pathways in some trypanosomatids have been partially characterized and the detection of polyisoprenoids in Leishmania such as ubiquinone, ergostatrienol and ergostadienol as well as of the enzyme HMG-CoA reductase have already been described (Ranganathan et al., Int. J. Parasitol. 25:279, 1995; Ginger et al., Biochem. J. 342:397, 1999; Montalvetti et al., Biochem. J. 349:27, 2000). However, in Leishmania, leucine and acetyl-coenzyme A are the substrates for the mevalonate pathway.In Trypanosoma brucei, cholesterol, ubiquinone and dolichol biosynthesis from [3H]mevalonate was demonstrated. Short chain dolichols with 11-12 isoprene units were found, compared to 15-23 unit dolichols found in mammals (Low et al., J. Biol. Chem. 266:19250, 1991). In species of Leishmania, the biosynthesis of these isoprenoids has not been fully characterized.Synthesis of dolichol and of the isoprenic chain of ubiquinone and the characterization of dolichol in L. amazonensis are being investigated by metabolic labelling of promastigotes with [2-14C] mevalonate. The analysis of the hexane fraction by HPLC allowed the identification of labelled dolichol of 11-12 isoprenic units. The same approach will allow us to verify the presence and characteristics of labelled ubiquinone. The study of isoprenoid biosynthesis in Leishmania is highly relevant since it could represent a target for the development of useful drugs for the treatment of leishmaniasis
