Follistatin in the mouse uterus during the first half of pregnancy (2003)

- Abstract: During the pregnancy in mammalian, endometrial fibroblasts differentiate into decidual cells. Biochemical and "in vitro" experiments made in rats, showed that decidual cells secrete some cytokines such as follistatin that are engaged on the cellular proliferation and differentiation, both are important events for the success of pregnancy. Follistatin regulates interactions between trophoblast-decidua and controls the secretion of prolactin. The aim of this work was to analyze by immunocytochemistry the distribution of follistatin in the mice uterus during pregnancy. Sites of implantation were collected from mice from 1 ½ to 11-½ day of pregnancy (dop). The samples were fixed, embedded in paraffin and sections were obtained. Sections were immunocytochemically stained for detection of follistatin and the reaction was developed with 3,3' -diaminobenzidine. Follistatin was first detected in the endometrial stroma on 3 ½ dop.On 4 ½ and 5 ½ dop, follistatin was present in the antimesometrial stroma (AS) except for a small region of decidualized cells around the implantation crypt. From 6 ½ until 8 ½ dop, e follistatin continue to be present in the AS and appeared in the mesometrial stroma (MS). On 9 ½ dop, follistatin remained exclusively in the nondecidualized stroma. On 10 ½ and 11 ½ dop, follistatin was no longer detected at the AS, however, follistatin was strong in MS, particularly, in the junctional zone of the uterus. Thepresent results showed that the expression of follistatin in the uterus is a regulated process that seems to be correlated with proliferation and differentiation of different cell populations of the uterus. This hypothesis is reinforced by results obtained from the thymidine triatiated incorporation in uterus, which showed a close correlation between radioactively labeled cells and follistatin immunoreactive cells
