Rat airway hypereactivity and levels of tnf seric and tecidual (2003)

- Abstract: Objetivo: The airway hyperreactivity (AH) is characterised by an exacerbated smooth muscle contractile response to different stimuli, wich can to cause airway obstruction such as those observed in acute respiratory distress syndrome (ARDS). The LPS induceexert their effects by release of several mediators including tumor necrosis factor (TNF). The TNF is considered one of more important trigger of acute inflammatory responses to LPS. In the present we investigated the effect of LPS on AH to methacholine and their relation with seric and tissue level of TNF. Métodos e Resultados: Methods: Male Wistar rats (200-250 g) were treated with LPS (Salmonella abortus; 5 mg/kg, iv). Non-treated rats were used as controls. Trachea were isolated 90 min or 48 h after LPS, mounted in an organ bath chamber (containing Krebs-Henseleit buffer under 95% O2/ 5% CO2) and the dose-response curves (DRCs) to methacholine (MCh) were constructed by recording isometric contraction force. The animals were treated with dexamethasone or clorpromazine 30 min before of LPS-intravenous.Before sacrifice of animals, the blood was collected to dosage of TNF in serum. In other experiments, trachea segments were incubated with LPS (100 ng/ml) during 90min or 48 h and the TNF level tissue was measured. Results : DRC to MCh were significantly shifted to the left 48 h after of LPS when compared with non-treated rats. The maximal response to MCh were increased in all times studiedafter LPS when compared with those from non-treated rats (control: 10.38±1.07 vs LPS 90 min: 18.57±1.61 g/100 mg of tissue; control: 10.4±1.07 vs LPS 48 h: 15.67±0.86). The maximal response to MCh 90 min or 48 h after LPS were significantly reduced by dexamethasone or clorpromazine. The TNF seric level was detected only 90 min after LPS, but 48 h after LPS it was not detectable. Differently, the TNF tissue level was dectable in both times. Conclusões: The AH (90 min) seem to have relation with the TNF detected in serum, but after 48 h its not seen. Possibly the airway hyperreactivity observed lately (48 h) is correlated with the local production (tecidual) of TNF
