The imunossupressant fk506 and its effects on spinal cord contusion (2003)

- Abstract: Objetivo: Substantial functional and morphological impairments after spinal cord lesion is promoted by secondary degeneration. Manipulation of inflammatory responses i.e. by means of imunossupressant FK506 may be of interest. Métodos e Resultados: Adult male rats were submitted to a moderate spinal cord injury by dropping a 10g rod (25mm distance) using the NYU Impactor (Lesion Group). A group of animals were submitted to a sham operation (Sham Group). Some animals from both groups received FK506 intramuscular injection (0.5 mg/kg) daily on first week and every 48 hours on second week, while other rats received saline solution in the same fashion. The animals of Lesion Group had their bladders emptied by method of Credé and its distension checked. Motor and sensory evaluations were made using BBB Score, Combined Behavioral Score (CBS) and computerized infrared motion sensor activity monitor. The rats were sacrificed 14 or 85 days after contusion and their spinal cords frozen and sectioned.) The Cresyl Violet stained sections were submitted to stereological analysis. Number of remaining neurons (RN), lesion volume (LV), total spinal cord volume (TV), lesion area (LA) and total spinal cord area (TA) were quantified. The bladder function of the animals with lesion plus FK506 returned to normal earlier than animals with lesion plus saline solution. The BBB Score showed significant difference between Sham and Lesion Groups and also between Lesion PlusFK506 Group and Lesion Plus Saline Group (p<0.001, Anova). The CBS showed differences between Lesion Plus FK506 Group and Lesion Plus Saline Group in some specific time points: 21, 28, 63 and 70 days post injury (p<0.001, Anova). The Infrared Motion Sensor demonstrated that animals treated with FK506 moved less until 28th day post injury. That pattern was inverted after this period. The lesion index (Chadi et al., Intern. J. Neuroscience, 111:137-16, 2001), which is calculated multiplying the number of remaining neurons by the lesion volume fraction (LV/LT), was not different in groups that received FK506. ) The same occurred with other morphological parameters analyzed like lesion volume and areal fraction (LA/TA). Conclusões: FK506 does not alter morphological parameters of lesioned spinal cord, however it seems to accelerate the recovery after spinal cord injury by improving bladder and sensory-motor recovery
