Effect of thyroid hormone on angiotensin type 1 receptor (at1) expression in cultured cardiac myocytes and fibroblasts (2003)

- Abstract: Objetivo: Renin-Angiotensin System (RAS) has been associated with cardiac hypertrophy induced by thyroid hormone. However, little is known regarding the regulation of AT1 receptor in cardiac cells after hyperthyroidism. Métodos e Resultados: Newborn rat ventricular cardiomyocytes and fibroblasts were obtained from 1 to 2 day old Wistar rats. After successive digestion of the tissue, cardiomyocytes and fibroblasts were separated using a Percoll gradient and pooled in DMEN enriched with Horse or Foetal Bovine Serum, respectively. 24 hours before the treatment, cells were maintained in serum free media. Cells were treated with crescent doses of T3 (1?M - 1µM) during 24 hours. After that total RNA was extracted from two cell types and AT1 expression analyzed using semiquantitative RT-PCR. Results: After treatment, cardiomyocytes showed a significative decrease on expression level of AT1, characterizing a dose-response relation, which showed the lowest expression level with 1 µM of T3.) On the other hand, the treatment with T3 in cardiac fibroblasts did not significantly change the expression level of AT1, independent of dose used. Conclusões: The effect of Thyroid hormone on decreasing AT1 receptors levels on cardiac myocytes may play an important regulatory role considering that both Angiotensin II and Thyroid Hormone induced cardiac hypertrophy
