- Abstract: Objetivo: Recently, a Lis1-interacting protein named NUDEL was shown to be essential for brain development and cortical organization. It belongs to a family of vertebrate coiled-coil proteins, which associated to other neuronal proteins, promotes nucleokinesis and axonal outgrowth. Any impairment affecting this association results in serious pathologies, such as lissencephaly and schizophrenia. In this study, we demonstrate, using immunologycal and enzymatic assays, that the NUDEL not only has identical amino acid sequence, but also displays the same enzymatic and immunological features of the brain endooligopeptidase A, isolated from the cytosol of rabbit brain in the early 70s. Our studies also show that the NUDEL-oligopeptidase activity is predominant in the central nervous tissue of the rat, while substantial activity was also detected in the cytosol of testes, skeletal muscle and liver.) Métodos e Resultados: In this study, we attempt to distinguish rNUDEL from rTOP (thimet oligopeptidase) and rNL (neurolisin) using a series of internally quenched fluorescence substrates derived from bradykinin. The results showed that, for most amino acid substitutions, the changes in the specificity of the rat rEOPA were similar to those observed for the rTOP and the rNL. However, NUDEL could be unambiguously identified in tissue extracts, using aNUDEL, thus allowing distinguishing NUDEL from TOP and NL. Conclusões: We concluded that NUDEL displays the same enzymaticand immunological features of the brain endooligopeptidase A
