Molecular characterization of bovine rotavirus from São Paulo state, Brazil (2002)

- Abstract: Group A bovine rotaviruses (BoRV-A) are among the enteropathogenic agents frequently associated with neonatal diarrhea in calves. Serotype classification of group A rotavirus is based on two differents capsid proteins, VP4 and VP7, which characterize P and G serotypes, respectively. Field surveys demonstrated that, so far, G6 and G10 are the most prevalent G serotypes in calves. The most widespread bovine rotavirus P genotypes are P[1], P[5] and P[11]. Eleven bovine stool specimens were collected from animals with and without diarrhea from São Paulo state. In a previous research, all of the samples were screened for rotavirus by enzyme immunoassay (EIARA-FIOCRUZ) and PAGE. Four of these samples were isolated and propagated in MA-104 and CaCo2 cells, with characteristic cytopathic effect. Additionally, all samples were submitted to the subgroup and serological characterization by using an enzyme immunoassay with monoclonal antibodies (MAb Elisa). In the present study, molecular characterization of these eleven samples was performed using reverse transcription (RT) followed by polimerase chain reaction (PCR) with specific primers for common animal and human G and P genotypes. G genotype results showed five samples with G10 specificity, two with G6 and one with G2 specificity. By serological characterization with Mabs specific for human and animal serotypes of rotavirus, only two of these samples were characterized as G10For the P type, all samples werecharacterized: 5 samples were P[11] and mixtures of P[4]+P[11], P[8]+P[11], P[1]+P[4], P[5]+P[11] and P[4]+P[5]+P[11] were found in other samples. RT-PCR of two cultivated samples with P[5]+P[11] mixture showed only P[5] specificity, suggesting that cells culture selected one of the strains present in these samples. These results, specially the G[2]P[4] strain, characteristic of human rotavirus, suggests interspecies transmission of rotavirus, and contributes to a better understanding of the epidemiology of bovine and human rotaviruses in Brazil
