Diversity of G2 serotypes/genotypes of human rotavirus in São Paulo, Brazil (2002)

- Abstract: Human rotavirus A (HuRV-A) is an important agent of acute gastroenteritis of young children. Three viral proteins are used for characterization of RV-A: VP6 classifies rotavirus into subgroups, VP7 into G serotypes and VP4 into P serotypes/genotypes. We used serological and/or molecular technics for RV-A characterization in São Paulo, Brazil. From 158 stool samples from children with ages between 0 and 5 years, collected from March/94 to September/95, we identified 41 samples with G2 specificity by serotyping with monoclonal antibodies or genotyping by RT-PCR. In 19 samples, G2 serotypes were in agreement with results found by genotyping. In 10 samples, G2 serotype/genotype was found in mixture with other G specificities. From 19 samples with G2 serotype/genotype, 3 had long eletropherotype and subgroup II specificity: 2 were P[8] and one, P[4][8][6]. Ten samples showed short electropherotype, 9 of which was subgroup I and one was subgroup non I-non II. From this, 4 samples were P[4], 3 were P[6], two were P[8] and one samples was negative for P genotyping. Six samples showed a patterns with segment 10 in the usual position and segment 11 positioned very close to it (close electropherotype); all close samples were subgroup I and P[4]. These results shows the diversity of G2 HuRV-A in São Paulo city. These results contributes to a better characterization of rotavirus strains infecting childrens in Brazil and are very important if any vaccination program is intended
