Detection and molecular caracterization of G and P genotypes of human rotavirus from São Paulo, Brazil (2002)

- Abstract: Infantile diarrhea is responsible for many pediatric hospitalizations and for high mortality in developing countries. Rotavirus is the main pathogen associated with diarrhea in young children. Fecal samples (236) were collected from December/1995 to November/1999 from 3 hospitals and one ambulatory in São Paulo city, Brazil. Samples were submitted to the enzyme immunoassay for rotavirus and adenovirus (EIARA-FIOCRUZ) and polyacrylamide gel eletrophoresis (PAGE). Results showed 44 (18,6%) samples positive for rotavirus by EIARA and 35(14,8%) by PAGE. Samples were then analyzed by genotyping RT-PCR. Two sets of primers were used, one for G genotypes (G1, G2, G3, G4 and G9) and other for P genotypes (P[8], P[9], P[4], P[10] and P[6]). Preliminary results for G genotype characterization showed 6 G1 samples, 5 G2, 2 G4 and 5 G9 samples. Five samples showed mixed genotypes: one sample was characterized as G1+G4+G9, 2 as G1+G4 and 2 as G1+G2+G4. For P genotype, results are: 5 P[4] samples, 7 P[8] and 4 samples were characterized as mixtures of P[4]+P[8] genotypes. These results confirms the emergence of G9 genotype, detected in São Paulo since 1996 and shows the value of the study of rotavirus genotypes, with important applications for vaccine strategies
