- Abstract: Since its isolation in 1957, Respiratory Syncytial Virus (RSV) has been considered the major pathogen of respiratory tract of kids and babies. That is an important cause of severe respiratory disease in adults, immunocompromissed patients and premature babies, especially those with chronic pulmonary or cardiac disease. Two large antigenic subgroups of RSV were described, designed A and B. The surface glycoproteins, F and G, are the principal targets for the host's immune response against RSV infections. Antibodies against F and G proteins would have different functions, preventing fusion of cells (anti-F) and blocking the first contact of the virus with the cell (anti-G). The vacinal proposals that are being tested in human volunteers are derived from attenuated strains. Another possibility to prevent RSV infection is a DNA-based vaccine expressing the viral envelope glycoproteins F and G. We draw specific oligonucleotides to sub-clone F and G genes from the RSV genome using RT-PCR technique into the plasmid pShuttle. The mammalian expression cassette in pShuttle consists of the human cytomegalovirus immediate early promoter/enhancer, a multiple cloning site, and the bovine growth hormone polyadenylation signal. The vector backbone also possesses the pUC origin (pUC ori) and a kanamycin resistance gene (Kanr) for propagation and selection in E. coli
