- Abstract: Several studies propose a role for NF-kB as a signal in the brain during neurodegeneration. In addition, recent evidences suggest that this nuclear factor has a role in glial and neuronal function, including synaptic transmission, development, and plasticity. The aim of this study was to investigate whether acute stress or chronic stress (14 days) can regulate the NF-kappaB activation in the CNS. Adult male Wistar rats were allocated into four groups: control (no-stress), acute stress predictable stress and unpredictable stress. To evaluate the influence of the glucocorticoids in these experiments, another 4 groups of animals were treated with metyrapone (200mg/kg/day) during the stress period. Cerebellum, hippocampus, and striatum were isolated from animals killed 24h after the last stress. Gel mobility shift assay was used to measure changes in NF-kB activation with treatments. The unpredictable stress induced a decrease in NF-kB activity in all brain regions analyzed. Metyrapone treatment blocked this response only in the cerebellum, suggesting that other intracellular pathways may be involved in this adaptative process
