Regulation of insulin-induced Tyrosine-Phosphorylation of IRS-1, IR-2, and IRS-3, in adipose tissue of two distinct animal models of insulin resistance: aging and glucocorticoid excess in vivo (2002)

- Abstract: The insulin receptor substrates molecules, IRS-1, IRS-2, and IRS-3 are key mediators in insulin signaling in the adipose tissue. IRS-1 and IRS-2 are wide expressed and located mainly in the low density microsome fraction while IRS-3, specific of adipose tissue, is located mainly in the plasma membrane fraction. These tree substrates are mediators of insulin-induced PI3-kinase activation. Although the IRS-3-null mice showed no difference from wild-type in body weight, in blood glucose level, and in fasted and fed insulin levels, the regulation of IRS-3 in conditions that show insulin resistance are not known. In the present study, we examined the phosphorylation state of insulin receptor (IR), IRS-1/2 and IRS-3 in adipose tissue of aging rats as well as dexamethasone treated rats. In adipose tissue from 12-month-old rats, insulin-induced tyrosine phosphorylation in anti-phosphotyrosine immunoprecipitates were reduced to 65±9% for IR, to 50±8% for both IRS-1 and IRS-2, and to 78±2% for IRS-3 compared to 2-month-old controls rats. In a same fashion way, the 2-month-old dexamethasone treated rats had a reduction to 47±3% for IR, to 52±3% for both IRS-1 and IRS-2, and to 68±9% for IRS-3. These pattern of reduced insulin-induced tyrosine phosphorylation of IRS proteins in the adipose tissue of two models that exhibit hyperinsulinemia and obesity may have a role in the insulin resistance observed in these models
