Tipificação genômica de mycoplasma Gallisepticum por amplificação polimórfica randômica de DNA (RAPD) (2002)

- Abstract: Mycoplasma gallisepticum (MG) é o agente causador da doença crônica respiratória das aves e sinusite infecciosa dos perus. Amplificação polimórfica randômica (RAPD) é um método que tem sido usado no estudo da heterogenicidade entre cepas de MG. O RAPD utiliza-se de primers arbitrários para o reconhecimento de seqüências alvo do DNA. O perfil gerado no RAPD resulta do anelamento dos primers arbitrários escolhidos com seqüências seletivas e amplificações de regiões flanqueadas. No presente estudo foram analisados 27 primers utilizando ciclos diferentes no RAPD. A análise da relação genética entre as cepas de MG, incluindo cepas de referência e vacinais e nove isolados de campo, foram feitas por meio de modelos matemáticos. A extração do DNA cromossomal foi obtida pelo método de fervura. Os parâmetros de amplificação foram padronizados testando-se diferentes concentrações de DNA, Taq DNA polimerase, primers, MgCl2 e dNTP. Vinte e quatro primers de 10 pares de base e três primers longos (S10, M13 e M16) foram testados com três diferentes ciclos de amplificação. A eletroforese em gel de agarose foi corada em solução de brometo de etídeo, visualizada em UV, fotografada e analisada pelo Vilber Lourmat Photodocumentation System, que calculou o peso molecular de cada fragmento. A matriz de similaridade foi obtida com base no coeficiente de Jaccard`s. A relação genética entre as cepas foi resultado das médias (UPGMA) e os dendogramas originados por meio do RAPD com diferentesprimers e ciclos. Estes métodos permitem uma tipificação de cepas de MG para estudos epidemiológicos, considerando diferenças entre cepas vacinais e selvagens
