Coagulation in the spider Acanthoscurria gomesiana (2002)

- Abstract: Coagulation is a fundamental reaction in the arthropod hemostasis and immune response. This reaction in horseshoe crab is induced by bacteria and fungi endotoxins which activate a proteolitic cascade resulting in the conversion of the clottable protein (coagulogen) into a insoluble gel (coagulin). All cascade components are present in the hemocytes. In order to purify the spider coagulogen, the hemocyte proteins were fractionated in a size exclusion chromatography followed to coagulation assay with trypsin. The whole spider hemocyte extract was submitted to SDS-PAGE and Western blot using horseshoe crab coagulin antiserum (HCA). However, both assays did not indicate the presence of coagulogen in the hemocytes. Searching coagulation factors on 5760 randomly sequenced clones from a spider hemocyte cDNA library, sequences with high degree of similarity with all horseshoe crab coagulation factors were found, except for the coagulogen sequence. These results suggest that the spider coagulogen maybe present in the plasma instead of inside the hemocytes. The spider plasma proteins were fractionated by size exclusion chromatography and the collected fractions were submitted to SDS-PAGE and Western blot using HCA. One protein with molecular mass 19.1 kDa reacted with HCA. The final purification and characterization of this putative coagulogen is underway. The results so far obtained suggest some diferences between the horseshoe crab and spider coagulation
