Suppressive effect of Ascaris suum extract on immune response to ovalbumil is independent of IFN-G and IL-12 (2002)

- Abstract: Ascaris suum extract (Asc) down-regulates the immune response to ovalbumin (OA) as well as preferentially stimulates a Th2 response to its own components, which is responsible for the suppression of the OA-specific Th1 response. IL-4 plus IL-10-specific monoclonal antibody treatment of mice before immunization with OA + Asc abolishes this suppression. The immunosuppressive activity of Asc is due to high MW proteins (PI) that are obtained by gel filtration chromatography. Although PI stimulates IL-4 and IL-10 secretion, IgE and anaphylactic IgG1 antibodies against PI are only obtained in IL-12 or IFN-g-/-deprived mice. In this work, we investigated the role of IL-12 and IFN-g in the suppression exerted by PI on an unrelated immune response. IFN-g- or IL-12-deficient (IL-12-/- / IFN-g-/-) or wild-type mice were immunized with OA or OA + PI in complete Freund's adjuvant and challenged in the hind footpad with aggregated OA after 8 days. Immediate and delayed-type-hypersensitivity (DTH) reactions and antibody were compared in these mice. Hypersensitivity reactions and OA-specific IgE, IgG1 and IgG2a remained dampened in IL-12-/- or IFN-g-/- OA + PI-immunized mice. In agreement with previous results obtained in our lab, a diminished DTH reaction was obtained in IL-12-/-, OA-immunized mice, in contrast to an increased reaction in IFN-g-/- mice. These results suggest that IL-12 and IFN-g do not participate in the down-regulation of anti-OAresponse induced by PI, ever though they act differently in OA-specific DHT
