Immunological propierties and vaccine potential of flagellin expressed by attenuated Salmonella enterica strains (2002)

- Abstract: Salmonella flagellin genetically fused to B-cell-stimulatory epitopes delivered parenterally is capable of eliciting antibody (Ab) responses against both flagellin and the fused antigen. Attempts to deliver this vaccine by mucosal routes (oral or nasal) showed that no significant systemic or local Ab response was elicited either to flagellin or to the fused epitope . In this work, we evaluated cytokine responses elicited in BALB/c mice orally immunized with three 1010 c.f.u. doses of S. enterica sv Dublin SL5928 strain transformed with plasmid pFLAII, encoding na hybrid flagellin (FLAII) fused with T-cell epitope contained within a 15 amino-acids peptide derived from CFA/I fimbriae of enterotoxigenic E. coli (ETEC). Spleen cells from immunized mice produced elevated IFN-gamma levels against both flagellin and the heterologous epitope. In contrast, no significant IL-4 response against either antigen was detected. Further experiments indicated that the flagellin- and CFA/I-specific IFN-gamma responses were mainly dependent on CD4+ cells. These results indicate that orally introduced Salmonella-hybrid-flagellin vaccines induce strong IFN-gamma responses against flagellin and genetically fused heterologous T-cell epitopes. Thus, this type of vaccines may represent a valuable oral vaccine approach to pathogens controlled by cell-mediated immune responses
