- Abstract: Objetivo: Atherosclerosis is a chronic inflammatory process in which components of the immune system (i.e., antibodies and T-cells) play a pivotal role. As moLDL has been implicaded in the atherosclerotic process the aim of this work was to test chromatographically separated (Size exclusion) subfractions of moLDL (oxidized with copper ) for their reactivity to autoantibodies in plasma of HC that is known to have a higher risk for coronary heart diseases (CHD) and compare the pattern of reactivity to that of plasma autoantibodies found in NC. Métodos e Resultados: moLDL obtained by oxidation of LDL (20 uM CuSO4, in vitro) was subfractionated using FPLC and Superdex 200 column (Pharmacia, Sweden) and a diode-array detector (Shimadzu, Japan). The total oxLDL and the protein peak fractions were assayed by ELISA for their reactivity with sera from HC (n=30, cholesterol > 250 mg/dL) and NC (n=30, cholesterol < 200 mg/dL). We could obtained 4 principal new peaks from moLDL. Of these peaks ( low molecular weight) one showed different reactivity with HC plasma than with NC. Conclusões: We can conclude that HC is a condition where we can detect a different spectrum of autoantibody reactivity against moLDL compared to NC. We think that this approach can be a useful methodology in the search for specific antigens in modified LDL involved in the formation of auto-antibodies in patients that are prone to develop atherosclerosis
