ESTS from hemocytes of the spider Acanthoschurria gomesiana: preliminary analysis (2002)

- Abstract: Spiders, as other invertebrates, have an efficient immune system to protect them against pathogens. Hemocytes play an important role in this mechanism, either by phagocytosis or by the production of antimicrobial molecules. To facilitate studies on the immune system of spiders, we generated a normalized cDNA library using mRNA extracted from hemocytes of adult spiders. At the present, 5760 clones have been partially sequenced. These sequences were processed to identify 4889 high-quality ESTs (Expressed Sequenced Tags) with an average length of 758 bp. A total of 2893 transcripts (clusters) were identified with the assembling of the sequences using CAP3. From these, 2056 clusters contained only one sequence (singlets) and 837 were a consensus of redundant sequences. The rate of new transcripts per sequenced clone was 60% and the number of clusters with more than 20 sequences was low, indicating a low redundance of the cDNA library. Sequence similarity search against nr database and open reading frame analysis suggest that 20% of the transcripts are putative new genes. Annotation using Gene Ontology terms and identification of immune related transcripts are in progress. This collection of spider hemocyte ESTs is an important tool to study the immune system of this organism using a functional genomic approach
