Effect creatine supplementation upon skeletal muscle wasting during immobilazation (2002)

- Abstract: Hypokinesia induces changes in various parameters of the skeletal muscle, such as, mass and creatine content. Thus the aim of this study was to investigate the effect of oral creatine supplementation upon muscle wasting during immobilization. Materials and Methods: 12 Wistar rats were divided in 2 groups: Control (C) and Supplemented with creatine (Cre). The left leg (I) was immobilized in shortening position and the contra-lateral leg remained lose (CL). The creatine supplementation protocol utilized was 0.5 g.kg-1(BW).day-1 for 7 days. After the sacrifice, hindlimb muscles were removed and muscle mass was determined (mg/g). The evaluation of lean mass development (g/cm) was performed by Dual Energy X-Ray Absortometry (DEXA - Norland©). (ver tabela no livro de resumos)Discussion: The immobilization procedure decreased relative wet weight of gastrocnemius (Gas) and soleus in both groups. No statistical difference was observed between Cre-I and I. However, relative muscle mass of Cre-I leg was slightly higher (soleus - 13% (p=0.15) and gastrocnemius - 6% (p=0.2) than I leg. The lean mass also suffered a marked decrease by the immobilization in both groups. Creatine supplementation failed to attenuate lean mass wasting (Cre-I x I - 7% - p=0.5). On the other hand, the lean mass development in Cre-CL was 36% greater than CL. In conclusion, creatine supplementation seems to play a major role in muscle mass gain during normal levels of muscular activity ratherthan disuse. e-mail: msaoki@usp.br
