- Abstract: Objetivo: Energy homeostasis is accomplished through a highly integrated and redundant neurohumoral system. Critical elements of this control system are hormones secreted in proportion to body adiposity, including insulin and leptin, and the hypothalamus targets upon which they act. Insulin and leptin have overlapping effects on the control of energy homeostasis, but the basis of this synergism is not known. Insulin signals through a receptor tyrosine kinase (IR) that phosphorylates and activates the docking proteins IRS, whereas the leptin receptor (OBR) and its associated protein tyrosine kinase JAK2 mediate phosphorylation and activation of the transcription factor STAT3. Métodos e Resultados: In the present study we have examined the activation of leptin and insulin signaling molecules in the hypothalamus of normal rats in vivo by immunoprecipitation, immunoblotting and EMSA. Male Wistar rats were studied after chronic implantation of an intracerebroventricular catheter into the third ventricle. Here we present a basis for the integration of leptin and insulin signals in the hypothalamus. Insulin induces OBR phosphorylation, which in presence of leptin promotes an augment in the interaction between STAT3 and the leptin receptor. Insulin also increases the leptin- induced phosphorylation of STAT3 and its activation. Conclusões: The results indicate that insulin modulates leptin signal- transduction pathway, and may provide a molecular basis for thecoordinated effects of insulin and leptin in feeding behavior and weight control
