Sodium channel agonists release nitric oxide (no) from rabbit isolated corpus cavernosum (2001)

- Abstract: Objetivo: Objectives: Tityus serrulatus scorpion venom (TSV) relaxes rabbit corpus cavernosum (RbCC) by a mechanism involving Na+ channel activation in nitrergic fibres (Teixeira et al., Br. J. Pharmacol., 123: 435 - 442, 1998). In this study, we have investigated the ability of Na+ channel agonists such as g toxin (Tg), veratridine (VTD) and brevetoxin-3 (PbTx-3) to release NO from RbCC. Métodos e Resultados: Methods: Male New Zealand rabbits (2 - 2.5 kg) were anaesthetized with SagatalÒ (40 mg/ kg, i.v.). The RbCC was dissected and mounted in a organ bath. Data were recorded in a MacLab computer system. Results: Addition of the NO synthesis inhibitor L-NAME (100 µM) increased the tone of RbCC tissues and prevented the relaxations induced by Tg (1 µM, n=3), VTD (30 µM, n=5) and PbTx-3 (100 mM, n=3). Subsequent addition of L-arginine (1 mM) reversed the increased tone and restored the relaxations. The soluble guanylate cyclase inhibitor ODQ (10 µM) increased the tone of RbCC tissues and prevented the relaxations evoked by VTD (n=6) and Tg (n=4). The Nasup>+ channel blocker tetrodotoxin (100 nM; n=3-6) abolished Tg (1 µM) and PbTx-3 (100 nM)-induced RbCC relaxations. When added during the relaxing response, tetrodotoxin promoted an immediate return to the basal line. Conclusões: VTD, PbTx-3 and Tg relaxes isolated RbCC through the release of NO from NANC nitrergic fibres
