Expression of neuronal isoform of nitric oxide synthase in spinal neurons of neonatal rats after sciatic nerve transection (2001)

- Abstract: Objetivo: The neuronal isoform of nitric oxide synthase (nNOS), a NADPH- dependent diaphorase, has been considered to play a role in motoneuron death induced by sciatic nerve transection of rodents during the first postnatal week. Correlation was found between motoneuron loss and presence of NADPH-d positive motoneurons. We examined the morphology and distribution of lumbar nNOS positive cells on the fifth day after neonatal rat sciatic nerve transection when the proportion of NADPH-d positive neurons were found to be maximal. We also examined the same features in the others spinal levels. Métodos e Resultados: Sciatic nerve of 8 neonatal rats (P2) was transected. At P7 the spinal cord was dissected. Cross sections were processed for nNOS immunohistochemistry and Nissl stain. Immunoreactivity to nNOS was examined in neurons in all spinal levels. We did not observe differences in immunostaining of lumbar neurons between operated and control sides. There was no morphological correspondence between lumbar motoneurons identified and counted in Nissl-stained material 9) and nNOS positive neurons in adjacent sections.?15; lesioned=61?(control=151 Densest staining was seen in laminae I-IV, from cervical through sacral levels. At thoracic and sacral levels, lamina VII contained groups of intensely reacting neurons linked by bundles of cell processes to numerous nNOS positive neurons situated around the central canal. Conclusões: Using immunohistochemistry instead ofhistochemistry for diaphorase activity, 5 days after axotomy, we found no evidence that alterations of nNOS expression underlie motoneuron demise following sciatic transection in neonatal rats
