Molecular identification of G[9]P[8] genotypes of human rotavirus in São Paulo, Brazil (2001)

- Abstract: Rotaviruses have been considered the most important cause of gastroenteritis in young children around the world. The two capsid proteins, VP7 and VP4 are used for molecular characterization of genotypes G and P, respectively. Fecal samples collected in São Paulo, Brazil from December 1995 to November 1999, were screened for rotavirus with EIARA Fiocruz Kit. From a total of 236 samples, 44(18,6%) were positive for rotavirus in EIARA.Objectives: The bjective of this work was to chacterization this samples by RT-PCR. Methods and Results: Preliminary results of genotypes G characterization by RT-PCR using a pool of primers for G1, G2, G3, G4 and G9 in 11 positive samples resulted in 3 samples with G[9] specificity. These results were confirmed by using the G[9] specific primer 9T-9B alone. These 11 samples were also tested by RT-PCR for genotype P characterization and the same 3 G[9] samples were characterized as P[8]. Conclusions: The unusual G[9] genotype detected for the first time in São Paulo, Brazil suggests, as many others studies worldwide, that genotype G[9] viruses may be widely distributed and more important than previously recognized
