The effect of diabetes mellitus on the lymphatic activity (2001)

- Abstract: Introduction: Multiple abnormalities in cellular and vascular function are described in experimental diabetes mellitus. On the other hand, the abnormalities in the lymphatic system activity have not yet been characterized. Thus, the effects of alloxan-induced diabetes on the lymph flow and lymphocyte traffic was investigated. Methods: Diabetes mellitus was induced in male Wistar rats with an injection of 40 mg/kg iv alloxan, and the animals were studied 10 days after. The presence of diabetes was verified by blood glucose concentrations > 200 mg/dl. The animals were anesthetized with chloral hydrate (40 mg/kg ip). A sylastic catheter was placed in the thoracic duct just below the diaphragm. The animals were then comfortably restrained in Bollman cages. Lymph was collected for 24 hours. Results: Lymph flow and lymphocyte traffic in the thoracic duct were greatly increased in diabetic compared with control animals (25.93 ± 2.66 vs 11.75 ± 0.94 mL and 589.61 ± 78.58 vs 250.87 ± 39.22 x 106 lymphocytes, p < 0.05). On the other hand, the number of blood lymphocytes did not change significantly in either group (7,159 ±1.269 vs 7,698 ± 771 lymphocytes/mm3). Conclusion: These results suggest that not only vascular function is altered in diabetic animals but the lymphatic system activity is also altered. Further studies are thus warranted to evaluate the consequences of increased transport of lymph and lymphocytes from tissues to the blood circulation in diabetes
