Vencular reactivity to vasodilators in diabetes mellitus: influence of the polyol pathway (2001)

- Abstract: Introduction: Hyperglycemia - induced acceleration of polyol pathway mediates a number of metabolic derangements that result in cellular dysfunction in diabetes mellitus. Goals: The present experiments were undertaken to verify the influence of inhibition of polyol pathway by minalrestat, on the decreased responses of mesenteric microvessels to mediators of inflammation in alloxan-diabetic rats. Methods: Diabetes mellitus was induced in 170-190g male Wistar rats with an i.v. injection of 40 mg/Kg alloxan. Control rats were sham-injected with physiologic saline. Minalrestat treatment (10 mg/Kg/d/30d) started 3 days after alloxan injection. In anaesthetized animals the mesentery was exteriorized for microscopic observation using a closed TV circuit coupled to the microscope. Venular diameter changes were measured using an image splitting device. The effect of topical application of acetylcholine (300 mg/ml), sodium nitroprusside (1mg/ml), bradykinin (3 mg/ml), histamine (50 mg/ml) and PAF (6 mg/ml) were analyzed. Results: The decreased responses of venules to bradykinin, histamine and PAF in diabetic rats (4.62 ± 0.59; 4.58 ± 0.55; 4.77 ± 0.46, respectively) in comparison to control (14.23 ± 1.45; 11.37 ± 0.68; 10.42 ± 1.22) was corrected by minalrestat (11.1 ± 0.86; 12.78 ± 0.91; 10.03 ± 0.42, respectively). Similar responses to acetylcholine and sodium nitroprusside were obtained in venules of diabetic (10.30 ± 1.2; 9.38 ± 1.0), control (10.10 ± 1.3; 11.40 ± 0.9) andtreated diabetic rats (11.85 ± 1.6; 10.31 ± 0.6). Conclusions: Polyol pathway, exacerbated in diabetes mellitus, might be involved in the impaired microvessel reactivity
