- Abstract: Objetivo: One of the effects of thyroid hormone is converting slow twicht fiber towards fast twitch fiber. Considering that the metabolism of fast fiber type is predominantly glycolytic in contrast to the oxidative of slow fiber type, our study aimed to demonstrate the effect of thyroid hormone upon myoglobin (Mb) mRNA and Protein expression, as well as Citrate Synthase (CS) and Succinate Dehydrogenase (SDH) activities which would provide an indirect evidence of oxidative capacity of the muscles.Métodos e Resultados: MB gene expression, CS and SDH activities of S and EDL muscles were studied in intact, thyroidectomized and T3- treated (25 ug/ 100g, BW, ip, 15 day-treatment) rats.The fiber type composition of S muscle was also evaluated. In the S muscle, the T3 treatment increased the Mb mRNA and protein expression, the CS and SDH activity. These changes occurred in parallel to the expected increase in type II (fast) and decrease in type I (slow)-fibers in S muscle. In the hypothyroid state, the Mb mRNA was decreased, while the Mb expression and CS activity tended to decrease. On the EDL, the alterations were milder than those of S muscle in both thyroid states.Conclusões: Mb gene expression is induced by T3, which, in parallel to the enhancement of Krebs Cycle enzymes activities, provide an additional evidence that thyroid hormone increases the aerobic potential of skeletal muscles, at the same time that it increases the speed of musclecontraction
