Immunorectivyti in omentum-associated lymphomyeloid tissue of mice infected with paracoccidioides brasiliensis (2006)

- Resumo: Introduction and Objectives: Distinct patterns of lesions have been observed in omental lymphomyeloid tissue of susceptible (B10.A) and resistant (A/J) mice to Paracoccidioides brasiliensis (Pb) infection. IFN-ã and osteopontin (OPN) are implicated in cellular immunity and in granulomatous responses, however, their participation in fungal granulomas has been poorly investigated. Our aim was to study the pattern of tissue lesions correlating to in situ expression of IFN-ã and OPN in a murine model of Pb infection. Methods and Results: Conventional staining and immunohistochemical methods were used to analyse the granuloma formation and to detect IFN-ã and OPN expression in omentum of B10.A and A/J mice after i.p. infection with the highly virulent Pb strain, Pb18. IFN-ã positive (+) lymphocytes were detected at the periphery of granulomas in both mouse strains and semiquantitative analysis (mean ± SEM, cells/mm2, n=3) showed a significant increase in + cells in compact granulomas of A/J mice at 120 days post-infection (dpi) compared to 15 dpi (6.40 ± 0.76 vs. 1.44 ± 0.54).The number of + cells in A/J granulomas at 120 dpi was significantly higher than that observed in loose and multifocal granulomas of B10.A at the same time point (2.47 ± 0.18). On the other hand, OPN was strongly + in macrophages and in multinucleated giant cells (MGCs) localized mainly in the center of the lesions. Intense OPN + extracellular matrix (ECM)was only detected at 120 dpi, when an increased expression in macrophages and in MGCs around Pb yeasts, mainly localized in the center of active granulomas was found. Significantly more (mean ± SEM, n=3) OPN + cells in B10.A (0.54 ± 0.01) than in A/J (0.24 ± 0.01) at 15 dpi were counted, and a significant increase of + cells of A/J at 120 dpi (0.50 ± 0.06) was found. Higher degree of OPN + ECM was detected at 120 dpi than at 15 dpi in both B10.A (0.11 ± 0.02 vs. 0.03 ± 0) and A/J (0.16 ± 0.02 vs. 0.01 ± 0.01). Conclusions: This work suggests that the omental lymphomyeloid tissues are sites of intense immunoreactivity, as shown by expression of IFN-ã and OPN in active granulomas. These components may have important roles in the tissue response during Pb infection.
