- Resumo: Introduction: The modifications of lipid and apolipoprotein B components of LDL drive the atherosclerosis process. These modified components can interact and activate macrophages. These properties depend on modifications. These can range from minimal to extensive. The extension/degree of modification and its subsequent Immunobiological properties remains hither to not completely understood. Objectives: In this study we compared the minimally oxidative modification (mmLDL) and highly oxidated LDL (HoxLDL) induced by iron or copper respectively, with the objective to detect differences with respect to biochemistry and immunobiological properties. Methods: LDL was prepared by ultracentrifugation. The oxLDL was prepared by addition of CuSO4 (20 µM 18h, 37 ºC) or FeSO4 (1 µM for 48 h RT). One part was labeled by FITC. To analyze the biochemical properties (size, charge) we used size exclusion (Superdex 200R) and anions exchange chromatography (Resource-Q, NaCl gradient 0- 500 mM) respectively using a FPLC system. Then we compared the uptake by THP-1 (using the FITC labeled). The change of expression of CD36, CD11b, CD32 (FcãRII) and CD128 (CXCR2) was analyze by FACS.
