The effects of chronic no inhibition on the contractile responses of isolated rat detrusor smooth muscle by electrical field stimulation (EFS) (2005)

- Resumo: Objetivo: The detrusor contraction involves two components: the parasympathetic nervous system and an atropine resistant, nonadrenergic noncholinergic system (NANC) mediated mainly by ATP. NO is produced in the nitrergic neurons, but its role in modulating the detrusor responses is not clear. The aim of this work was to investigate the effects of chronic NO inhibition on the rat detrusor contractile responses during EFS. Métodos e Resultados: Methods: Wistar male rats were treated orally with L-NAME (20mg/rat/day) for 30 days. Age-matched control animals received tap water alone. The activity of NO synthase activity (NOS) assay was determined. The detrusor contractile responses during EFS were carried in the absence and presence of four autonomic blockers (1 bM), namely atropine, guanethidine, phentolamine and propanolol to obtain the NANC responses. The long-term L-NAME treatment induced a marked arterial hypertension, and reduced by 64% the calcium-dependent NOS activity in the isolated rat detrusor smooth muscle. Tetrodotoxin (1bM) abolished the EFS-induced rat detrusor smooth muscle contractions, whereas in vitro addition of L-NAME (100 bM) did not alter these contractile responses. Long-term treatment with L-NAME increased by approximately 60% the EFS (1-30 Hz)- induced contractions in the detrusor preparations. In preparations treated with the autonomic blockers in order to obtain the NANC excitatory responses, long-term L-NAME treatment increased by 10 %(P<0.05) the EFS-induced contractions. The potentiation by L-NAME was higher (P<0.01) in parasympathetic-mediated contractions in comparison with the NANC responses. Conclusões: Our findings suggest that NO exerts a modulatory role in both parasympathetic and NANC excitatory fibers
