Basic fibroblast growth factor immunoreactivity in neurons and satellite cells of the myenteric plexus of the rodent ileum (2005)

- Resumo: Objective: The enteric nervous system consists of a number of interconnected networks of neuronal cell bodies and fibers as well as satellite cells, the enteric glia. Basic fibroblast growth factor (bFGF, FGF-2) is a mitogen for a variety of mesodermal and neuroectodermal-derived cells and its presence has been described in many tissues. The present work employs immunohistochemistry to analyze bFGF immunoreactivity in neurons and glial cells in the rat and mouse iliac enteric plexus. Methods and Results: Adult male pathogen-free Wistar rats and C57BL/6J mice from the Institute of Biomedical Sciences (Sao Paulo, Brazil) were processed for immunohistochemistry; the distal ileum was opened and its myenteric plexus was processed as whole-mount preparations. The membranes were immunostained for visualization of FGF-2 and glial fibrillary acidic protein (GFAP). FGF-2 immunoreactivity was observed in the majority of the neurons and satellite cells of the enteric ganglia of the ileum. GFAP-immunoreactive enteric glial cells and their processes were present in the iliac enteric plexus surrounding neuronal cell bodies and axons. A dense net of GFAP-immunoreactive processes was seen in the ganglia and connecting strands of the myenteric plexus. The two-color immunofluorescence method revealed bFGF immunoreactivity in GFAPpositive enteric glial cells. Conclusion: The results suggest that the presence of FGF-2 in neuron and satellite cells of the myenteric plexus of therodent ileum may be related to autocrine and paracrine actions of the molecule in the enteric nervous system
