Sequence analysis of VP4 gene of porcine rotavirus strains displaying human P[6] genotype specificity (2003)

- Abstract: Porcine rotaviruses are a significant cause of diarrhea in swine farms worldwide. A total of 59 porcine rotavirus samples were previously characterized for G and P genotypes by reverse transcription-polymerase chain reaction (RT-PCR) with specific primers for porcine, bovine and human rotavirus genotypes. Common porcine G serotypes and P genotypes G3, G4, G5, P[6] and P[7] and atypical serotypes G9 and G10 were detected. Five samples from different farms in the Paraná State, Brazil showed P[6, M37]+P[6,Gott] specificity. As the P6 (M37 like) genotype is normally associated with viruses causing infection in humans, these viruses were further characterized by partial sequencing of their VP4 gene. Results showed 87,0 - 91.2% nucleotide identity to P[6] M37 human strain and only 78.4 82.6% identity with P[6] Gottfried porcine strain. Samples showed less than 73.6% identity when compared with sequences representing all other P genotypes. By Blast analysis, all five samples produced significant alignments with human P[6] rotavirus strains. Our results suggests that P[6] rotavirus strains occurring in pig farms in Southern Brazil are closely related to human strains, indicating the possibility of emergence of new porcine strains through reassortment between human and pig rotaviruses
