- Abstract: Introduction: Experimental autoimmune uveitis (EAU) is an organ-specific, T cell-mediated disease that targets the posterior pole of the eye. EAU can be induced in rodents and nonhuman primates by immunization with retinal antigens such as interphotoreceptor retinoid-binding proteins (IRBP). Alternative immunotherapeutic approaches to autoimmune uveitis are a major target of vision research, because the treatments available have numerous and severe side effects. Using The EAU model we have studied one such immunotherapeutic alternative. Methods: B10.RIII mice were immunized with 100 mug of IRBP emulsified in CFA and were given 0,5 mug of Pertussis toxin (PTX) in 0,1ml i.p. as an additional adjuvant. Group I mice were treated with 10 mg/dose of antagonist of the VLA-4 in the phase efferent (14, 16 and 18). Group II received 10 mg/dose of antagonist of the VLA-4 but in the phase afferent (2, 4 and 6) and group III mice were untreated (control).Draining lymph nodes (LN) were collected 21 days after immunization and the cells were cultured to evaluate the proliferative responses to IRBP. Disease severity was evaluated by fundoscopy at day 14 and by histological examination of the enucleated eyes at day 21. Delayed-type hypersensitivity (DTH) responses to IRBP was calculated as the difference between the thickness of the IRBP-injected foot pad and the PBS-injected foot pad 24 and 48 hours after the injection of antigen. Results: Ourresults demonstrated that the treatment with antagonist of VLA-4 inhibit the EAU development and suppress the proliferative and delayed-type hypersensitivity responses. The effect of antagonist of the VLA-4 on EAU are under investigation. Conclusion: Treatment with antagonist of the VLA-4 can modulate the development of EAU and block the cell migration to eye
