Location of DNA repair genes in relation to gene transfer islands (2003)

- Resumo: Horizontal gene transfer (HGT) is a collective name for processes that permit the exchange of DNA among organisms of different species, even between distant evolutionary lineages; thus, the most reliable method for detection of HGT events is phylogenetic analysis and recognition of unusual patterns of evolution. HGT of a given gene is strongly correlated with its gene function: "informational genes" (those participating in transcription, translation, replication and DNA repair) are less prone to be horizontally transferred than "operational genes" (those involved in housekeeping functions). In this work, we describe the location of DNA repair genes from two sequenced Xanthomonas genomes, Xanthomonas axonopodis pv citri and Xanthomonas campestris pv campestris, in relation to potentially "gene transfer islands" found in these genomes. The search for horizontally transferred genes was made by analysis of unexpected best matches, via BlastP algorithm, and the repair genes candidates to HGT were analyzed in an evolutionary manner, via phylogenetic reconstruction.For nearly one hundred repair genes of both Xanthomonas, distributed along seven repair pathways, almost thirty percent of DNA repair genes are located in the given "gene transfer islands" or in the vicinity. From these, we focused our phylogenetic analysis in genes belonging to direct repair, nucleotide excision repair, base excision repair and mismatch repair pathways. Theproximity of Xanthomonas genes with orthologs of unrelated organisms to define possible horizontally transferred genes is a widely accepted assumption, and in this way we identified 10 repair genes potentially introduced via HGT in both Xanthomonas, including phr, ada, alkA, ogt and uvr genes
