Glucocorticoid exacerbation of LPS-induced activation of NF-kB in pre-frontal cortex (FCx) by cronic unpredictable stress (USt) (2003)

- Resumo: Glucocorticoids (GCs) are stress hormones that can compromise the ability of neurons to survive a variety of insults. The transcription factor NF-kB regulates numerous genes that play important roles in inter and intracellular signaling, cellular stress response, and degeneration. The aim of this study was to investigate the role of GCs in LPS-induced activation of NF-kB FCx, using USt (14 days) as experimental model. Adult male Wistar rats (n=6) were exposed to USt for 14 days and in the 15th day, both control (C) and USt animals rats were treated with saline or LPS (1mg/kg, i.v.). Two hours later, all rats were decapitated and the hypothalamus (HYP), hippocampus (HIPC) and FCx removed. Influence of GCs was studied by treating rats with mifepristone (RU486), a GC receptor antagonist (50 mg/kg, i.p.), 30 min before LPS injection. NF-kB activity was studied by gel shift assays.When compared to C, USt induced a further increase in LPS-activation of NF-kB in FCx but not in HYP and HIPC. Supershift assay indicated that the major DNA-protein complex altered by LPS corresponded to p50/p65 heterodimer. RU-486 treatment was able to block this response induced by LPS in FCx, without changing LPS effects in HYP and HIPC after USt. Our results suggest that Ust induced pro-infammatory effect mediated by glucocorticoid in FCx
