Effects of in vivo exposure to lips on the reactivity of rat airway smooth muscles (2003)

- Resumo: Objetivo: LPS is a potential inductor of the respiratory failure observed in acute respiratory distress syndrome. The airway hyperreactivity is characterized by increased smooth muscle contractile response to different stimuli wich can eventually cause airway obstruction. The mechanisms of hyperreactivity induction caused by LPS are not well understood. In the present study we investigated the direct and indirect effects of LPS on the cholinergic stimulation of rat isolated trachea. Métodos e Resultados: METHODS:Trachea rings of Wistar rats (n=7 for each group)) treated in vivo (5mg/kg,iv) during 90 min, 6, 24, 36 or 48 h with LPS (Salmonella abortus equi) were set up for the measurement of isometric contractions in 10-ml organ baths containing Krebs-Henseleit solution (37ºC, 95% O2, 5% CO2).) RESULTS:In in vivo results obtained indicating that the response to MCh, 90 min (LPS : 18.5 ± 1.6 g vs control: 10.4 ± 2.8 g), 6 h (LPS: 13.68±1.27 vs control: 10.38±1.07), after exposure to LPS caused increase of maximal response respectively when compared with control groups. The augment of maximal response (LPS 48 h: 15.67±0.66 vs control: 10.38±1.07) and the shift to the left of dose response curve was observed 48 h after LPS. In times of 24 or 36 h after LPS, was not observed increase of maximal response but the curve was shift to the left (24 h: 10-7 M, 10-6 M, 10-5 M) and (36 h: 10-6 M, 10-5 M). Conclusões: The airway smooth muscle reactivity and sensibility oncholinergic stimulation (MCh) are time in vivo dependents of exposure to LPS. Suggested that LPS can exert indirect influences (via mediators release) on airway smooth muscle, altering sensibility to cholinergic stimulation
