Computer assisted nyu impactor system to promote different degrees of contusion lesion in the rat spinal cord (2003)

- Abstract: Objetivo: Different degrees of partial contuse experimental lesion in the rat spinal cord may be necessary in the study regarding local degenerative/regenerative events. Métodos e Resultados: Adult male Wistar rats were submitted to different degrees of spinal cord contusion lesion to demonstrate experimental models of local injury by means of a NYU Impactor System. The spinous process and the vertebral lamina at T9-10 level were removed to expose a circular region of the dura. To promote the lesions, a 10g rod was dropped to the dorsal surface of the exposed spinal cord from distances of 12.5 mm, 25 mm or 50 mm, which characterize three intensities of lesion: slight (SL), moderate (MD) or severe (SV), respectively. The Impactor registered different variables: impact velocity (ImpV), compression distance (Cd), compression time (Ct), and compresion rate (Cr=Cd/Ct). Impact velocity was measured from a regression line fit to the rod trajectory during the 2 msec before it contacts the spinal cord.Compression distance was the maximum distance below the baseline. Compression time was the time (msec) required for the rod to reach the maximum compression distance. Compression rate (Cr) was the calculated parameter (Cd/Ct). Impact curve was calculated by the system and the parameters of the trauma, like impact velocity, cord compression distance and cord compression rate were obtained in order to demonstrate the severity of the trauma. Thefollowing results were obtained with the three intensities of the lesion: Cd [SL= 1.957 ± 0.097; MD= 2.582 ± 0.0257; SV= 3.302 ± 0.0326, p=0.01]; ImpV [SL= 3707.2 ± 92.59; MD= 2793.34 ± 60.75; SV= 2062.63 ± 50.14, p=0.02] and Cr [SL= 3.64x10-4 ± 036x10-4; MD= 5.73x10-4 ± 0.35x10-4; SV= 7,9. 10-4 ± 0,23.10-4, p=0.0001]. Conclusões: The results indicated that the computer assisted Impactor System is a device capable to reproduce different degrees of spinal cord contusion in rats and to provide measurements with precision
