The effect of T. cruzi infection over EAE development (2002)

- Resumo: AIM: to study how intracellular parasite infections can modulate the development of autoimmune diseases. Methods: IL-10 or iNOS knockout (KO) mice in the C57Bl/6 background and their matching wild type (WT) controls were immunized with 100 ug of MOG 35-55 peptide emulsified vol/vol in CFA enriched with 400 ug of M. bovis. Each mouse received 100 ng of Pertussis toxin intravenously at the same day of immunization and 2 days after, as an additional adjuvant. Groups of mice were also infected with 1000 blood forms of T. cruzi Y strain concomitantly to immunization or 21 days later. After the immunization/infection, the clinical score of EAE was followed and lymph node cells (LNC) or spleen cells (SC) cultures were performed on days 7th, 14th or 21st to evaluate the proliferative and cytokine responses against the MOG peptide as well as the expression of CD95, CD95L, CD28 and CD152 molecules. Results: The infection with T. cruzi completely blocked the initial EAE development and controlled an ongoing disease. Cells from immunized/infected mice had an increased expression of CD95, CD95L, and CD152, suppressed lymphoproliferative responses against MOG peptide and higher levels of IL-10 and NO production. The IL-10 or iNOS KO mice reverts the protective effect of T. cruzi infection over EAE development. Conclusion: The intracellular parasite infections can modulate de development of autoimmune diseases and the elucidation of these control mechanisms could cooperate to thedevelopment of new therapies against human autoimmunities
