Molecular characterization by gene sequence of atypical genotype G[3] bovine rotavirus sample from the State of Goiás, Brazil (2001)

- Resumo: Objectives: Rotaviruses are the main cause of viral gastroenteritis on infants, young children and many other animal species worldwide. These viruses have a genome consisting of 11 segments of dsRNA each one coding for at least one protein. Rotaviruses genes coding for the two structural proteins located in the outer capsid (VP4 and VP7) are used for molecular characterization of G and P genotypes, by reverse transcription followed by polymerase chain reaction (RT-PCR). The purpose of this study was to characterize some atypical G[3] bovine rotavirus samples by gene sequence. Methods and Results: Four bovine rotavirus samples from Piracanjuba, Goiás, Brazil were previously characterized by RT-PCR, with aET3 primer, as genotype G[3], not yet described for bovine samples. One sample, ICB828, had its VP7 gene sequenced in an ABI-Prism 377 DNA sequencer, with Big Dye kit. Preliminary results showed that sequence of this sample has a homology of 91-93% with bovine G[6] NCDV and UK strains and less than 73% homology with several G[3] strains. Nucleotide sequence analysis of ICB828 showed that, at the G3-specific primer binding position (689 to 709) only 5 mismatches were present. In this position ICB828 has only one mismatch with G6 NCDV strain and 100% homology with G6 UK strain, leading to the conclusion that this G3 primer reacts with bovine G6 strains. To confirm this hypothesis, three standard bovine strains, NCDV G[6], UK G[6] and B223 G[10] were submitted to aRT-PCR using the set of primers for characterization of human rotavirus samples. All of them reacted with aET3 primer. Conclusion: These results suggest that the aET3 G[3] primer should be used with caution for characterization of bovine rotavirus samples
