Transcription-coupled and global DNA repair gene in sugar cana (2001)

- Resumo: Objectives: To search DNA repair related genes in the Sugar Cane EST Genome Project (ONSA Consortion, FAPESP). Methods and Results: The analysis was performed mainly by comparison with BLAST (Basic Local Alignment Search Tool) of genes identified in other eukaryotes (Arabidopsis thaliana, human and yeast) or even bacteria (E.coli). Among the genes analyzed, a few were selected for their participation in transcription-coupled and global genome nucleotide excision repair of damaged DNA. The ESTs reads corresponding to these genes were chosen by their insert sizes, and three candidates for full lengths were completely sequenced, until finishing quality was achieved. CSA and CSB are two human genes that are part of the transcriptional-coupled repair apparatus. Deficiency in these genes lead to the hereditary disease Cockayne Syndrome. Both CSA and CSB homologues from sugar cane were sequenced. CSA clone is missing a few aminoacids at the N-terminal and CSB clone is complete. Curiously, CSB clone has the A.thaliana homologue annotated as a TATA box binding protein associated factor. The third clone chosen is a XPC homologue, responsible for the human disease xeroderma pigmentosum. Unfortunately, the sugar cane clones are not full lengths. Conclusions: Because the genes are not complete we are trying to obtain the 5'-end of such genes and confirm their functions in plants
